The evolutionary success of bacteria depends greatly on their capacity to continually generate phenotypic diversity. Structured environments are particularly favorable for diversification because of attenuated clonal interference, which renders selective sweeps nearly impossible and enhances opportunities for adaptive radiation. We examined at the microscale level the emergence and the spatial and temporal dynamics of phenotypic diversity and their underlying causes in Escherichia coli colonies. An important dynamic heterogeneity in the growth, metabolic activity, morphology, gene expression patterns, stress response induction, and death patterns among cells within colonies was observed. Water is vital for plant performance and survival. Its scarcity, induced by a seasonal decline in soil water availability or an increase of evaporative demand, can cause failures of the water conducting system. An adequate tolerance to drought and the ability to acclimate to changing hydraulic conditions are important features for the survival of long-lived woody plants in dry environments. In this study we examine secondary growth and xylem anatomical acclimation of 6 year old saplings of three European oak species (Quercus robur, Q. petraea, Q. pubescens) during the third consecutive year of exposure to soil drought and/or air warming (from 2007 to 2009). Chromosomes that determine sex are predicted to evolve differently than autosomes: a lack of recombination on one of the two sex chromosomes is predicted to allow an accumulation of deleterious alleles that eventually leads to reduced functionality and potential physical degradation of the nonrecombining chromosome. Because these changes should occur at an elevated evolutionary rate, it is difficult to find appropriate species in which to test these evolutionary predictions. The unique genetic sex-determining mechanism of the crustacean Eulimnadia texana prevents major chromosome degeneration because of expression of both ‘proto-sex’ (i.e. early stage of development) chromosomes in homozygous form (ZZ and WW). Herein, we exploit this unique genetic system to examine the predicted accumulation of deleterious alleles by comparing both homogametic sexual types to their heterogametic counterpart. We report differences in crossing over in a sex-linked region in the ZW hermaphrodites (∼ 3%) relative to the ZZ males (∼ 21%), indicative of cross-over suppression in the ZW hermaphrodites. Additionally, we report that both ZZ and WW genotypes have reduced fitness relative to ZW hermaphrodites, which is consistent with the prediction of harboured recessive mutations embedded on both the Z and the W chromosomes. These results suggest that the proto-sex chromosomes in E. texana accumulate recessive deleterious alleles. We hypothesize that recessive deleterious alleles of large effect cannot accumulate because of expression in both ZZ and WW individuals, keeping both chromosomes from losing significant function.

In wild populations, individuals are regularly exposed to a wide range of pathogens. In this context, organisms must elicit and regulate effective immune responses to protect their health while avoiding immunopathology. However, most of our knowledge about the function and dynamics of immune responses comes from laboratory studies performed on inbred mice in highly controlled environments with limited exposure to infection. Natural populations, on the other hand, exhibit wide genetic and environmental diversity. We argue that now is the time for immunology to be taken into the wild. The goal of 'wild immunology' is to link immune phenotype with host fitness in natural environments. Coevolutionary arms races between males and females have equipped both sexes with mutually manipulative and defensive adaptations. These adaptations function to benefit individual reproductive interests at the cost of the reproductive interests of opposite-sex mates, and arise from evolutionary dynamics such as parental investment (unequal reproductive costs between the sexes) and sexual selection (unequal access to opposite-sex mates). Individuals use these adaptations to hijack others' reproductive systems, psychological states, and behaviors--essentially using other individuals as extended phenotypes of themselves. Such extended phenotypic manipulation of sexual rivals and opposite-sex mates is enacted by humans with the aid of hormones, pheromones, neurotransmitters, emotions, language, mind-altering substances, social institutions, technologies, and ideologies. •Analyses of plant tolerance in response to different modes of herbivory are essential to an understanding of plant defense evolution, yet are still scarce. Allocation costs and trade-offs between tolerance and plant chemical defenses may influence genetic variation for tolerance. However, variation in defenses also occurs for the presence or absence of discrete chemical structures; yet, the effects of intraspecific polymorphisms on tolerance to multiple herbivores have not been evaluated.•Here, in a glasshouse experiment, we investigated the variation for tolerance to different types of herbivore damage, and direct allocation costs, in 10 genotypes of Boechera stricta (Brassicaceae), a wild relative of Arabidopsis, with contrasting foliar glucosinolate chemical structures (methionine-derived glucosinolates vs glucosinolates derived from branched-chain amino acids).•We found significant genetic variation for tolerance to different types of herbivore. Structural variations in the glucosinolate profile did not influence tolerance to damage, but predicted plant fitness. The vast majority of animal species display range fidelity, a space-use behaviour enhancing familiarity with local habitat features. While the fitness benefits of this behaviour have been demonstrated in a variety of taxa, some species or populations rather display infidelity, displacing their home range over time. Others, such as many ungulate species, show seasonal adjustments in their range fidelity to accommodate changes in the dominance of limiting factors or in the distribution of resources. Few empirical studies have explored the adaptive value of seasonal adjustments in range fidelity. Mitochondria are essential multifunctional organelles whose metabolic functions, biogenesis, and maintenance are controlled through genetic interactions between mitochondrial and nuclear genomes. In natural populations, mitochondrial efficiencies may be impacted by epistatic interactions between naturally segregating genome variants. The extent that mitochondrial-nuclear epistasis contributes to the phenotypic variation present in nature is unknown. We have systematically replaced mitochondrial DNAs in a collection of divergent Saccharomyces cerevisiae yeast isolates and quantified the effects on growth rates in a variety of environments. We found that mitochondrial-nuclear interactions significantly affected growth rates and explained a substantial proportion of the phenotypic variances under some environmental conditions. Mawi DNA Technologies has developed the iSWAB sample collection system and Copan is producing FLOQSwabs™ (hDNA free, certified free of human amplifiable DNA, DNase and RNase free). The combination of the Mawi iSWAB system and Copan FLOQSwabs™ hDNA free integrates into a non-invasive sample collection and preservation technology that delivers between 10 - 30µg of DNA with < 1% bacterial contamination and can be used to easily collect samples from all population segments including infants and elderly. The Mawi iSWAB system captures buccal cells collected by Copan FLOQSwabs™ hDNA free at the point of collection, so there is no need to process swabs in the lab. Samples are stable at room temperature for >1 year. Samples collected by this system are ideal for downstream microarray or NGS platforms. Applications include genetic analysis, epigenetics, forensics, and animal testing. Although the system has been validated with different swab types, it was demonstrated by several customers that the Copan FLOQSwabs™ are compatible with Mawi iSWAB, and have been yielding a larger amount of nucleic acids.

We hypothesized that changes in southeastern Bering Sea foraging conditions for black-legged kittiwakes (Rissa tridactyla) have caused shifts in habitat use with direct implications for population trends. To test this, we compared at-sea distribution, breeding performance, and nutritional stress of kittiwakes in three years (2008–2010) at two sites in the Pribilof Islands, where the population has either declined (St. Paul) or remained stable (St. George). Foraging conditions were assessed from changes in (1) bird diets, (2) the biomass and distribution of juvenile pollock (Theragra chalcogramma) in 2008 and 2009, and (3) eddy kinetic energy (EKE; considered to be a proxy for oceanic prey availability).
